Polymersomes are interesting in that they have a unique combination of advantageous properties compared to liposomes, dendrimers, micelles and solid particles, for use in nanomedicine and controlled release applications. These properties include:

  • Site-specific targetting possible by polymersome surface derivation

  • Possible to dial in a range of bioresorbtion rates using FDA approved materials

By proper tailoring of the amphiphilic blockcopolymers that make up the shell of polymersomes in aqueous environments, the following parameters can be controlled: Form, size, bioresorbtion rate, stability, shell thickness and permeability, surface derivation anchors.

Site-specific targeting
Polymersome surfaces can by proper choice of polymerization conditions be made with handles that enable derivation of surfaces with a controlled amount of functional groups. Popular examples for such functional groups are:

  • Aminogroups (Bioconjugation)
  • Aldehydegroups  (Bioconjugation)
  • Carbon double bonds (Hydrosilation)
  • Carbon triple bonds (Click-reaction)

To achieve site-specific targeting of polymersomes biological recognition motives can be attached to the polymersomes surface by either standard bioconjugation techniques, click-chemistry routes or other coupling methods.
